ISOLASI DAN KARAKTERISASI XILANASE DARI BACILUS CIRCULANS

Abstract

A research of islation and characterization of xylanses (1,4-β-D-xylan xylanohydroalse, EC 3.2.1.8) that suitable for pulp pre-bleaching has been conducted. Several bacillus has the ability to produce thermostable and alkalistable xylanse. The objective of this experiment was to isolated and charcterized xylanase from bacillus circulans. First, xylanase was isolated. The extracellular enzyme was partially purified by fractionation step using ammonium sulphate (20-40% saturation) and ion excchange chromatography DEAE-ToyoPEARL. Optimum pH and temperatureof the partically purified enzime were determined. Enzime purification using ion exchange chromatography showed an increase in spesific activy (805.48 U/mg) with 46.8 fold purification. Partially purified xylanase from B. Circulans exhibited favorable potential for pre-bleaching stage in pulp and paper industry because have some advantages in technical, economical and environmental aspect

Penelitian mengenai isolasi dan karakterisasi xilanase (1,4-β-D-xylan xylanohydroalse, EC 3.2.1.8) yang pengunaannya sesuai dengan kondisi proses pra pemutihan pulp telah dilakkan. Beberapa isolat bacillus diketahui mampu menghasilkan xilanase yang tahan pH alkali dan suhu tinggi. Dari penelitian ini xilanase dari bacillus circulans diisolasi dengan karakterisasi. Enzim ekstraselular yang diperoleh dimurnikan secara faksial dengan fraksinasi menggunakan ammonium sulfat (persen saturasi 20-40%) yang dilanjutkan dengan kromatografi penukar ion DEAE-toyoPEARL. Pada pH dan suhu optimum xylanase hasil pemurnian kemudian dikarakterisasi. Pemurnian enzim menggunakan kromatografi penukar ion menunjukan adanya peningkatan aktivitas enzim spesifik (805,48 U/mg) dengan kelipatan pemurnian 46,8 kali dibandingkan dengan ekstrak kasarnya. Xylanase hasil pemurnian persial ini diharapkan dapat digunakan untuk proses pra pemutihan pada industri pulp dan kertas karena memiliki beberapa keunggulan baik dari sisi teknis, ekonomi, danlingkungan.
